腺病毒D型探針法熒光定量PCR試劑盒實驗操作:
1. 模板制備（樣本制備區(qū)）
建議使用配套水生動物病害基因組 DNA/RNA 提取試劑盒系列產(chǎn)品，具體過程詳見產(chǎn)品說明書。
2．添加模板（模板添加區(qū)，放置于冰盒中進(jìn)行）
請于-20℃條件下保存，有效期 12 個月
取出所需測試數(shù)的已含有反應(yīng)液 A-TSV-I 的 PCR 管，將試劑*解凍，離心 30 秒，在管蓋上標(biāo)記管名（陰性對照管 NG、樣品 XX、陽性對照管 PG）。打開管蓋向各管管底分別加入 0.8μL B-I 及 0.2μL R-I，蓋上陰性對照管管蓋，其他各管分別沿管壁加入 2μL 模板，順序為待測樣品模板、PG-TSV-I，依次蓋好各管管蓋，離心 30 秒，立即進(jìn)行擴增反應(yīng)。
3. 擴增反應(yīng)（擴增及產(chǎn)物分析區(qū)）
①恒溫儀器 63℃條件下反應(yīng) 60 min。待儀器升溫至 63℃后，新建程序，設(shè)置實驗名稱及反應(yīng)時間，將步驟 2中離心后的 PCR 反應(yīng)管放入恒溫?zé)晒夥肿訖z測儀，點擊開始檢測。
②若使用熒光定量 PCR 儀，則熒光基團(tuán)選擇 FAM，淬滅基團(tuán)選擇 None，將 63℃ 15 s，63℃ 45 s 作為一個循環(huán)，于 63℃ 45 s 處收集熒光信號，60 個循環(huán)。其他儀器請參照儀器說明書進(jìn)行設(shè)置。
